  1. Sample Collection: DNA samples are collected from the individuals or biological samples under investigation. Common sample types include blood, saliva, semen, hair follicles, and tissue samples.
  2. DNA Extraction: The DNA is extracted from the collected samples using laboratory techniques designed to isolate and purify the DNA from other cellular material.
  3. PCR Amplification: Polymerase chain reaction (PCR) is used to amplify specific regions of the DNA that contain highly variable genetic markers, such as short tandem repeats (STRs) or variable number tandem repeats (VNTRs). These regions, also known as genetic loci, exhibit variations in the number of repeated DNA sequences between individuals.
  4. Fragment Analysis: The amplified DNA fragments are separated and analyzed using gel electrophoresis or capillary electrophoresis. This technique separates the DNA fragments based on their size, allowing for the visualization of the unique pattern of DNA fragments at each genetic locus.
  5. DNA Profiling: The pattern of DNA fragments obtained from each genetic locus is used to create a DNA profile or fingerprint for the individual. This profile consists of a series of peaks representing the sizes of the DNA fragments at each genetic locus.
  6. Comparison and Interpretation: The DNA profiles obtained from different samples are compared to determine whether they match. A match indicates that the DNA samples came from the same individual, while differences in the DNA profiles indicate different individuals.
